When cells are provided with the correct resources and conditions researchers can culture everything from bacteria to human tissue. When assessing the activity of cells researchers must identify or design methods to measure cell activity since most changes cannot be visualized. Often in medical research scientists aim to measure the impact of a particular treatment on cell behavior or survival. Taking absorbance readings for cultures is one approach that is often used to indirectly measure the number of living cells. Researchers start by plating the cells in different wells of a plate and then expose them to a compound called a tetrazolium salt. Tetrazolium salts are water soluble and nearly colorless when alone. However when the salt is exposed to electron acceptors like those associated with reducing enzymes in cells the salt reduces to form a brightly colored dye. This dye is harmless to the cells but colors the plating medium limiting the light that can pass through. From here researchers process the plates in a special machine called a plate reader which is a type of spectrophotometer that broadcasts light at a chosen wavelength through every well of the plate. A sensor in the device reads the wavelength of light that passes through each well and compares it to the transmitted wavelength to obtain an absorbance value for each well. The more dye produced the higher the absorbance value. By using the plate reader/spectrophotometer to measure absorbance in each well the number of active cells in each can be quantified according to the amount of dye produced.
